What is GHK-Cu?
GHK-Cu is a natural complex formed by the tripeptide Gly-His-Lys (glycine-histidine-lysine) bound to a copper ion (Cu²⁺). It was discovered in 1973 by Loren Pickart in human plasma, where its concentration decreases significantly with age: from ~200 ng/mL at age 20 to ~80 ng/mL at age 60.
This decline correlates with the reduction in wound-healing and tissue-renewal capacity observed during aging, positioning GHK-Cu as a “renewal peptide” of interest in anti-aging research.
Mechanism of action
GHK-Cu acts through multiple pathways:
Collagen and elastin synthesis:
- Stimulates fibroblasts to increase production of collagen types I, III, and VI.
- Increases synthesis of elastin and glycosaminoglycans (hyaluronate, dermatan sulfate).
- Activates matrix metalloproteinases (MMPs) to eliminate damaged collagen and replace it with new collagen — a tissue remodeling process.
Gene regulation (microarray data):
- A 2010 study (Pickart, Margolina) showed that GHK-Cu modulates the expression of 4,000+ genes: activates genes associated with tissue repair and suppresses inflammatory genes.
- Among the activated genes: those related to protein synthesis, mitochondrial metabolism, and tumor suppression.
Antioxidant and anti-inflammatory:
- The copper in the complex has antioxidant properties via activation of superoxide dismutase (SOD).
- Inhibits TNF-α and IL-1β, key pro-inflammatory cytokines.
Research applications
Skin (topical use — best documented):
- Wrinkle reduction and firmness: multiple double-blind studies show improvement vs. placebo.
- Chronic wound healing.
- Reduction of UV damage.
- Hair follicle regeneration (alopecia in animal models).
Systemic use (research):
- Internal wound healing.
- Pulmonary protection in COPD models.
- Neuroprotective effects in animal models.
GHK-Cu in cosmetics vs. injectable research
Topical application is the best documented and safest form. Cosmetic products with GHK-Cu have been on the market since the 1980s with an excellent safety profile.
Injectable use is only in research contexts and lacks approved clinical protocols.
