Retatrutide enters phase 3 with the TRIUMPH program
The design of TRIUMPH was published: four phase 3 trials of the triple GIP/GLP-1/glucagon agonist, with more than 5,800 participants, in weight, sleep apnea and knee osteoarthritis. It is a design paper, not a results paper; phase 3 efficacy is not yet reported.

A 2025 meta-analysis of 5 RCTs (706 patients) found that Tα1 significantly reduced extrapancreatic infections, abdominal infections, and inflammatory markers (CRP, APACHE II). The documented mechanism is immune regulation: Tα1 raised CD4+ counts and restored the CD4+/CD8+ ratio.

At 48 weeks, the dual glucagon/GLP-1 agonist resolved MASH without worsening fibrosis in 47 to 62% of the dose groups versus 14% with placebo. It is phase 2, with nausea (66%) and vomiting (41%) common; phase 3 confirmation is still pending.
