How to reconstitute a peptide

Verified step-by-step process. Every detail matters to preserve protein integrity.

Materiales necesarios

Before starting, gather: the lyophilized peptide vial, a bacteriostatic water (BAC water) vial, U-100 insulin syringes (1 ml or 0.5 ml), and 70% alcohol swabs.

Bacteriostatic water contains 0.9% benzyl alcohol as a preservative. This is what allows safe multiple-use from the vial.

Wait at least 10 seconds before inserting the needle. The alcohol needs to evaporate completely. Inserting the needle too early can introduce alcohol residue into the vial and damage the peptide.

Usa una jeringa de insulina U-100

U-100 insulin syringes are the smallest and most precise available. Recommended: 1 ml for sufficient range, or 0.5 ml for small doses.

In a U-100 syringe: 100 units = 1 ml. If your calculated dose above is, for example, 5 units, it means drawing to the 5 mark on the syringe.

Before drawing the BAC water, draw the same volume of aire que quieres extraer. Luego inserta la aguja en el vial invertido and push that air in.

This step equalizes the pressure inside the vial and lets the water flow easily into the syringe without needing to create excessive vacuum.

Between 1 ml and 3 ml is acceptable. More water makes micro-dosing easier. 2 ml gives a good balance of concentration and precision.

The impact of water against the glass slows it down: it reaches the peptide slowly without breaking its bonds. A direct stream onto the powder applies mechanical force that denatures the protein: irreversible damage.

Nunca sacudas el vial. Agitar crea espuma, introduce burbujas of air and creates stress at air-liquid interfaces where peptides denature and aggregate irreversibly.

Instead, gently roll the vial between your palms or make slow circles. This mixes the content without damaging the protein structure.

Let the vial rest at room temperature for 15–30 minutes, gently swirling occasionally. The solution should become completely clear, with no lumps or visible particles.

If after 30 minutes there are undissolved particles, it may indicate peptide degradation or solvent incompatibility. Do not use.

Aspirar la dosis correcta

Invert the reconstituted vial and insert the needle. Pull the plunger to the number of U-100 units indicated by the calculator above. Check that there are no air bubbles in the syringe.

If there are bubbles, hold the syringe with the needle up and tap gently with your finger to make them rise, then carefully expel the air before injecting.

Almacenar en refrigerador

Store the reconstituted vial in the refrigerator at 2–8°C, away from direct light. Bacteriostatic water maintains sterility for 28 días. Después de ese plazo descartar el vial.

Never freeze a reconstituted vial: ice crystals break the protein structure. Lyophilized (unreconstituted) peptides can be frozen for up to 24 months.
