What is Epithalon?
Epithalon is a synthetic tetrapeptide with the sequence Ala-Glu-Asp-Gly (alanine-glutamic acid-aspartic acid-glycine). It was synthesized by Prof. Vladimir Khavinson at the St. Petersburg Institute of Gerontology as an analog of the pineal gland extract known as epithalamin.
Khavinson investigated peptide bioregulators for decades in the Soviet Union, with the goal of “restoring” the endocrine function of organs that decline with age. The pineal gland (which produces melatonin) is one of the first organs to show functional deterioration during aging.
Mechanism: telomerase and cellular longevity
The most cited finding about epithalon is its ability to activate telomerase in human cells in vitro:
- Telomeres are the protective “caps” of chromosomes that shorten with each cell division.
- Telomere shortening is one of the molecular markers of cellular aging.
- Telomerase is the enzyme that can lengthen telomeres, but it is inactive in most adult somatic cells.
- A 2003 study (Annals of the NY Academy of Sciences) showed that epithalon induced telomerase activity in human cells in culture, resulting in telomere elongation and extension of the number of cell divisions.
Animal studies
The strongest data comes from murine models:
- Wistar rats: extension of mean lifespan by ~13% and maximum lifespan by ~25%.
- Effects on tumor incidence, endocrine and neurological function.
- Restoration of melatonin patterns in aged animals.
Important limitations
- The vast majority of research originates from a single group (Khavinson Institute) without independent replication published in high-impact Western journals.
- Human studies are small and lack rigorous control groups.
- Extrapolating telomerase activation in vitro to anti-aging effects in vivo in humans is speculative.
- Telomerase activation is an area of oncological research (cancer cells also have active telomerase).
