What is Argireline?
Argireline (INCI: Acetyl Hexapeptide-3 or Acetyl Hexapeptide-8) is a synthetic hexapeptide derived from the N-terminal end of SNAP-25, a protein involved in the SNARE complex that governs the docking of neurotransmitter vesicles at neuromuscular junctions.

Proposed mechanism of action
In in-vitro models, Argireline has been shown to competitively inhibit the formation of the SNARE complex by mimicking the N-terminal fragment of SNAP-25. This interference may reduce the calcium-triggered release of catecholamines, potentially softening repeated muscle contractions responsible for dynamic expression lines.
The effect is entirely local and reversible. Unlike botulinum toxin, it does not cleave SNAP-25 — it competes with it. This distinction is important for formulation claims and consumer communication.
What the research says
Human clinical evidence is limited and largely industry-sponsored. A 2002 study by Blanes-Mira et al. demonstrated inhibition of catecholamine secretion in chromaffin cells at concentrations of 100 µM. A small double-blind clinical trial reported a ~30% reduction in the depth of expression wrinkles after 30 days at 10% concentration.
Independent replication in larger populations is lacking. The cosmetic industry widely accepts Argireline as a functional ingredient based on this mechanistic plausibility and the available small-scale data.
